Non-Drive Side Large Nut
The large conical nut on the non-drive side of the spindle has come loose after 2 years of use. I’m just not sure how hard to tighten it back up - hand tight or should I get a large spanner on it to tighten it properly?

I think it's better with a spanner, and you have to block the cassette to tighten it, I think I have already done it once

It's better to do it by hand (is more confortable).
You have to use a wrench to tighten it while you hold the pulley:
